Antarctic Bacterial Environmental Genomic Efforts

In collaboration with Dr. Ed Delong, we have screened a large contig Antarctic environmental DNA library for bacterial diversity. Several clones with bacterial rDNA genes have been selected for complete sequencing. In order to sequence the large clones (40-50 kb) they had to be subcloned first. We used a strategy including nebulization followed by blunt ligation and cloning into E. coli, then picked clones to fill ten 96-well plates for each library.
